Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005591.4(MRE11):c.1592C>G (p.Ser531Cys), citing Ambry Variant Classification Scheme 2023: The p.S531C variant (also known as c.1592C>G), located in coding exon 14 of the MRE11A gene, results from a C to G substitution at nucleotide position 1592. The serine at codon 531 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
