Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.3944T>C (p.Ile1315Thr), citing Ambry Variant Classification Scheme 2023: The p.I1315T variant (also known as c.3944T>C), located in coding exon 24 of the CFTR gene, results from a T to C substitution at nucleotide position 3944. The isoleucine at codon 1315 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
