NM_000245.4(MET):c.3077A>G (p.Tyr1026Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y1044C variant (also known as c.3131A>G), located in coding exon 14 of the MET gene, results from an A to G substitution at nucleotide position 3131. The tyrosine at codon 1044 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
