Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.834A>T (p.Glu278Asp), citing Ambry Variant Classification Scheme 2023: The p.E278D variant (also known as c.834A>T), located in coding exon 7 of the CFTR gene, results from an A to T substitution at nucleotide position 834. The glutamic acid at codon 278 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
