NM_002382.5(MAX):c.362G>C (p.Ser121Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S121T variant (also known as c.362G>C), located in coding exon 5 of the MAX gene, results from a G to C substitution at nucleotide position 362. The serine at codon 121 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
