Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.1673A>T (p.His558Leu), citing Ambry Variant Classification Scheme 2023: The p.H558L variant (also known as c.1673A>T), located in coding exon 5 of the AXIN2 gene, results from an A to T substitution at nucleotide position 1673. The histidine at codon 558 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
