This variant lies in the IRF6 gene (transcript NM_006147.4) at coding-DNA position 82, where T is replaced by G; at the protein level this means replaces tryptophan at residue 28 with glycine — a missense variant. Submitter rationale: This sequence change replaces tryptophan with glycine at codon 28 of the IRF6 protein (p.Trp28Gly). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and glycine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with an IRF6-related disease.
