Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_144997.7(FLCN):c.1349A>G (p.His450Arg), citing Ambry Variant Classification Scheme 2023: The p.H450R variant (also known as c.1349A>G), located in coding exon 9 of the FLCN gene, results from an A to G substitution at nucleotide position 1349. The histidine at codon 450 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
