Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000143.4(FH):c.156A>G (p.Ile52Met), citing Ambry Variant Classification Scheme 2023: The p.I52M variant (also known as c.156A>G), located in coding exon 2 of the FH gene, results from an A to G substitution at nucleotide position 156. The isoleucine at codon 52 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
