Uncertain significance for Myoclonic dystonia 11 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003919.3(SGCE):c.232+5G>A, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SGCE gene (transcript NM_003919.3) at 5 bases into the intron immediately after coding-DNA position 232, where G is replaced by A. Submitter rationale: This sequence change falls in intron 2 of the SGCE gene. It does not directly change the encoded amino acid sequence of the SGCE protein, but it affects a nucleotide within the consensus splice donor site of the intron. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a SGCE-related disease. Nucleotide substitutions within the consensus splice site are relatively common causes of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of nucleotide changes on RNA splicing suggest that this variant may alter RNA splicing, but this prediction has not been confirmed by published transcriptional studies.
