Uncertain significance for Nephrolithiasis/nephrocalcinosis — the classification assigned by Ambry Genetics to NM_000388.4(CASR):c.308C>T (p.Thr103Ile), citing Ambry Variant Classification Scheme 2023: The p.T103I variant (also known as c.308C>T), located in coding exon 2 of the CASR gene, results from a C to T substitution at nucleotide position 308. The threonine at codon 103 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
