Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005228.5(EGFR):c.374A>G (p.Tyr125Cys), citing Ambry Variant Classification Scheme 2023: The p.Y125C variant (also known as c.374A>G), located in coding exon 3 of the EGFR gene, results from an A to G substitution at nucleotide position 374. The tyrosine at codon 125 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
