While this particular variant has not been reported in the literature, loss-of-function variants in TMEM127 are known to be pathogenic (PMID: 20154675, 21156949). This sequence change deletes 2 nucleotides from exon 2 of the TMEM127 mRNA (c.124_125delAC), causing a frameshift at codon 42. This creates a premature translational stop signal (p.Thr42Glyfs*65) and is expected to result in an absent or disrupted protein product.
