NM_177438.3(DICER1):c.4915T>C (p.Tyr1639His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y1639H variant (also known as c.4915T>C), located in coding exon 22 of the DICER1 gene, results from a T to C substitution at nucleotide position 4915. The tyrosine at codon 1639 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
