NM_177438.3(DICER1):c.3541A>T (p.Asn1181Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N1181Y variant (also known as c.3541A>T), located in coding exon 20 of the DICER1 gene, results from an A to T substitution at nucleotide position 3541. The asparagine at codon 1181 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
