NM_001232.4(CASQ2):c.1131_1133del (p.Glu377del) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the CASQ2 gene (transcript NM_001232.4) at coding-DNA position 1131 through coding-DNA position 1133, deleting 3 bases; at the protein level this means deletes glutamic acid at residue 377. Submitter rationale: The c.1131_1133delAGA variant (also known as p.E377del) is located in coding exon 11 of the CASQ2 gene. This variant results from an in-frame AGA deletion at nucleotide positions 1131 to 1133. This results in the in-frame deletion of a glutamic acid at codon 377. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
