The glycine at codon 122 is replaced by arginine, an amino acid with dissimilar properties. This variant was reported in individual(s) with features consistent with craniosynostosis (Topa A et al. Front Genet, 2023 Jan;14:1322462). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
