Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.778G>A (p.Asp260Asn), citing Ambry Variant Classification Scheme 2023: The p.D260N variant (also known as c.778G>A), located in coding exon 5 of the NOTCH1 gene, results from a G to A substitution at nucleotide position 778. The aspartic acid at codon 260 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
