Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.6596G>A (p.Cys2199Tyr), citing Ambry Variant Classification Scheme 2023: The p.C2191Y variant (also known as c.6572G>A), located in coding exon 39 of the FLNA gene, results from a G to A substitution at nucleotide position 6572. The cysteine at codon 2191 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
