Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.3854A>C (p.Asp1285Ala), citing Ambry Variant Classification Scheme 2023: The p.D1285A variant (also known as c.3854A>C), located in coding exon 31 of the FBN1 gene, results from an A to C substitution at nucleotide position 3854. The aspartic acid at codon 1285 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
