Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000071.3(CBS):c.203C>T (p.Thr68Ile), citing Ambry Variant Classification Scheme 2023: The p.T68I variant (also known as c.203C>T), located in coding exon 1 of the CBS gene, results from a C to T substitution at nucleotide position 203. The threonine at codon 68 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
