NM_178452.6(DNAAF1):c.115T>G (p.Cys39Gly) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C39G variant (also known as c.115T>G), located in coding exon 1 of the DNAAF1 gene, results from a T to G substitution at nucleotide position 115. The cysteine at codon 39 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
