This variant lies in the TAOK1 gene (transcript NM_020791.4) at the canonical splice donor site of the intron immediately after coding-DNA position 352, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.352+1G>A intronic variant consists of a G to A substitution one nucleotide after exon 5 (coding exon 4) of the TAOK1 gene. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant was determined to be de novo in at least one individual with features consistent with TAOK1-related neurodevelopmental disorder (Ambry internal data). This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site. Based on the available evidence, this alteration is classified as pathogenic.
