Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_003128.3(SPTBN1):c.227C>T (p.Thr76Ile), citing Ambry Variant Classification Scheme 2023: The c.227C>T (p.T76I) alteration is located in exon 3 (coding exon 2) of the SPTBN1 gene. This alteration results from a C to T substitution at nucleotide position 227, causing the threonine (T) at amino acid position 76 to be replaced by an isoleucine (I). Based on data from gnomAD, the T allele has an overall frequency of <0.001% (1/251464) total alleles studied. The highest observed frequency was 0.006% (1/16256) of African alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
