Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005585.5(SMAD6):c.715G>C (p.Val239Leu), citing Ambry Variant Classification Scheme 2023: The p.V239L variant (also known as c.715G>C), located in coding exon 1 of the SMAD6 gene, results from a G to C substitution at nucleotide position 715. The valine at codon 239 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
