This variant lies in the SMARCAL1 gene (transcript NM_014140.4) at coding-DNA position 2321, where C is replaced by A; at the protein level this means converts the codon for serine at residue 774 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Variant summary: SMARCAL1 c.2321C>A (p.Ser774X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant allele was found at a frequency of 8e-06 in 251442 control chromosomes (gnomAD). c.2321C>A has been observed in individuals affected with Schimke Immunoosseous Dysplasia (Boerkoel_2002). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
