Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005475.3(SH2B3):c.89G>C (p.Cys30Ser), citing Ambry Variant Classification Scheme 2023: The p.C30S variant (also known as c.89G>C), located in coding exon 1 of the SH2B3 gene, results from a G to C substitution at nucleotide position 89. The cysteine at codon 30 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
