Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001754.5(RUNX1):c.58+2dup, citing Ambry Variant Classification Scheme 2023: The c.58+2dupT intronic variant results from a duplication of one nucleotide at the +2 position in intron 1 of the RUNX1 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear.
