This variant lies in the EPM2A gene (transcript NM_005670.4) at coding-DNA position 640, where C is replaced by G; at the protein level this means replaces proline at residue 214 with alanine — a missense variant. Submitter rationale: This sequence change replaces proline with alanine at codon 214 of the EPM2A protein (p.Pro214Ala). The proline residue is highly conserved and there is a small physicochemical difference between proline and alanine. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a EPM2A-related disease.
