Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000059.4(BRCA2):c.2884_2908del (p.His962fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change deletes 25 nucleotides from exon 11 of the BRCA2 mRNA (c.2884_2908del), causing a frameshift at codon 962. This creates a premature translational stop signal (p.His962Ilefs*2) and is expected to result in an absent or disrupted protein product. While this particular variant has not been reported in the literature, loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). For these reasons, this variant has been classified as Pathogenic.
