Uncertain significance for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_41258467)_(41258556_?)dup, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross duplication of the genomic region encompassing exon 4 of the BRCA1 gene. While the exact position of the duplicated exon cannot be determined from this data, the duplicated copy of this region is likely in tandem and in-frame, therefore preserving the integrity of the reading frame. While this particular gross triplication has not been reported in the literature, exon-level duplications in BRCA1 have been reported in patients affected with hereditary breast/ovarian cancer (PMID: 15475941, 21327469). Experimental studies and prediction algorithms are not available for this variant, and the functional significance of this duplication is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
