It preserves the integrity of the reading frame. Deletion of exons 10-14 in BRCA1 has been reported in two families with hereditary breast and/or ovarian cancers (PMID: 22460208, 16793929). It is also known as deletion of exons 11-15 and c.671-?_4675+?del in the literature. The gross deletion of exons 10-14 of the BRCA1 gene removes 1335 amino acids located in important functional domains of the BRCA1 protein that are required for proper function of the BRCA1 protein as well as the interactions with RAD50, RAD51 and PALB2 (PMID: 25652403). For these reasons, this variant has been classified as Pathogenic.
