Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000135.4(FANCA):c.14G>C (p.Trp5Ser), citing Ambry Variant Classification Scheme 2023: The p.W5S variant (also known as c.14G>C), located in coding exon 1 of the FANCA gene, results from a G to C substitution at nucleotide position 14. The tryptophan at codon 5 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
