NM_000051.4(ATM):c.700G>C (p.Ala234Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A234P variant (also known as c.700G>C), located in coding exon 6 of the ATM gene, results from a G to C substitution at nucleotide position 700. The alanine at codon 234 is replaced by proline, an amino acid with highly similar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
