Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_016222.4(DDX41):c.322A>G (p.Lys108Glu), citing Ambry Variant Classification Scheme 2023: The p.K108E variant (also known as c.322A>G), located in coding exon 4 of the DDX41 gene, results from an A to G substitution at nucleotide position 322. The lysine at codon 108 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
