Pathogenic for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.7145dup (p.Asn2383fs), citing Ambry Variant Classification Scheme 2023: The c.7082dupT pathogenic mutation, located in coding exon 47 of the NF1 gene, results from a duplication of T at nucleotide position 7082, causing a translational frameshift with a predicted alternate stop codon (p.N2362Qfs*3). This variant was reported as heterozygous in individual(s) with features consistent with Neurofibromatosis type 1 (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
