Pathogenic for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.3895A>T (p.Lys1299Ter), citing Ambry Variant Classification Scheme 2023: The p.K1299* variant (also known as c.3895A>T), located in coding exon 29 of the NF1 gene, results from an A to T substitution at nucleotide position 3895. This changes the amino acid from a lysine to a stop codon within coding exon 29. This variant was reported in individual(s) with features consistent with Neurofibromatosis type 1 (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
