Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.3770T>C (p.Leu1257Pro), citing Ambry Variant Classification Scheme 2023: The p.L1257P variant (also known as c.3770T>C), located in coding exon 28 of the NF1 gene, results from a T to C substitution at nucleotide position 3770. The leucine at codon 1257 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
