Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.79C>G (p.Gln27Glu), citing Ambry Variant Classification Scheme 2023: The p.Q27E variant (also known as c.79C>G), located in coding exon 2 of the NF1 gene, results from a C to G substitution at nucleotide position 79. The glutamine at codon 27 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
