NM_001267550.2(TTN):c.66770-1G>A was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the TTN gene (transcript NM_001267550.2) at the canonical splice acceptor site of the intron immediately before coding-DNA position 66770, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.39575-1G>A intronic variant results from a G to A substitution one nucleotide upstream from coding exon 144 of the TTN gene. This exon is located in the A-band region of the N2-B isoform of the titin protein and is constitutively expressed in TTN transcripts (percent spliced in or PSI 100%). Alterations that disrupt the canonical splice site are expected to result in aberrant splicing. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site. A resulting transcript is predicted to be in-frame and is not expected to trigger nonsense-mediated mRNAdecay; although, direct evidence is unavailable. This nucleotide position is highly conserved in available vertebrate species. Based on the available evidence, the clinical significance of this variant remains unclear.
