This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 12826, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 4276 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The p.Q3913* variant (also known as c.11737C>T), located in coding exon 44 of the TTN gene, results from a C to T substitution at nucleotide position 11737. This changes the amino acid from a glutamine to a stop codon within coding exon 44. This exon is located in the I-band region of the N2-B isoform of the titin protein and is constitutively expressed in TTN transcripts (percent spliced in or PSI 100%). This variant was reported in individual(s) with features consistent with dilated cardiomyopathy (Ambry internal data).
