NM_017636.4(TRPM4):c.1986C>G (p.Asp662Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D662E variant (also known as c.1986C>G), located in coding exon 14 of the TRPM4 gene, results from a C to G substitution at nucleotide position 1986. The aspartic acid at codon 662 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
