Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_024334.3(TMEM43):c.777C>G (p.His259Gln), citing Ambry Variant Classification Scheme 2023: The p.H259Q variant (also known as c.777C>G), located in coding exon 9 of the TMEM43 gene, results from a C to G substitution at nucleotide position 777. The histidine at codon 259 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
