Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_152594.3(SPRED1):c.134T>C (p.Phe45Ser), citing Ambry Variant Classification Scheme 2023: The p.F45S variant (also known as c.134T>C), located in coding exon 2 of the SPRED1 gene, results from a T to C substitution at nucleotide position 134. The phenylalanine at codon 45 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
