NM_152594.3(SPRED1):c.1030C>G (p.His344Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H344D variant (also known as c.1030C>G), located in coding exon 7 of the SPRED1 gene, results from a C to G substitution at nucleotide position 1030. The histidine at codon 344 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
