Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_016938.5(EFEMP2):c.1289C>T (p.Ser430Phe), citing Ambry Variant Classification Scheme 2023: The p.S430F variant (also known as c.1289C>T), located in coding exon 10 of the EFEMP2 gene, results from a C to T substitution at nucleotide position 1289. The serine at codon 430 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
