Pathogenic for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.1890_1891dup (p.Gln631fs), citing Ambry Variant Classification Scheme 2023: The c.1890_1891dupCC (p.Q631Pfs*6) alteration, located in exon 14 (coding exon 14) of the DSP gene, consists of a duplication of CC at position 1890, causing a translational frameshift with a predicted alternate stop codon after 6 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. for DSP-related cardiomyopathy; however, its clinical significance for DSP-related cardiomyopathy with woolly hair, keratoderma, and tooth agenesis is uncertain. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
