Pathogenic for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_024422.6(DSC2):c.509_512dup (p.Tyr171Ter), citing Ambry Variant Classification Scheme 2023: The c.509_512dupACTA (p.Y171*) alteration, located in coding exon 5 of the DSC2 gene, consists of a duplication of ACTA at position 509 to 512. This changes the amino acid from a tyrosine (Y) to a stop codon at amino acid position 171. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
