Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000784.4(CYP27A1):c.101C>A (p.Ala34Asp), citing Ambry Variant Classification Scheme 2023: The p.A34D variant (also known as c.101C>A), located in coding exon 1 of the CYP27A1 gene, results from a C to A substitution at nucleotide position 101. The alanine at codon 34 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
