NM_201596.3(CACNB2):c.544A>T (p.Met182Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M128L variant (also known as c.382A>T), located in coding exon 4 of the CACNB2 gene, results from an A to T substitution at nucleotide position 382. The methionine at codon 128 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
